Overview

60S ribosomal protein L31 is a structural protein encoded by the RPL31 gene and forms an integral component of the large ribosomal subunit (60S) within the cytoplasm, essential for catalyzing protein synthesis in eukaryotic cells[1][3][4]. RPL31 belongs to the L31E family of ribosomal proteins and facilitates both translation and pre-rRNA processing during ribosome maturation[2]. While RPL31 is not a conventional drug target, changes in its expression and genetic integrity have been implicated in various pathological processes, including familial adenomatous polyps, resistance mechanisms in advanced prostate cancer, and Diamond-Blackfan anemia[2][4]. Mutations or altered expression of RPL31 can disrupt ribosome biogenesis, affecting cell proliferation and survival, especially in rapidly dividing tissues. Interaction partners such as BRCA1 have been reported, implying involvement in broader regulatory networks[1][3]. As a core ribosomal element, its disruption is more associated with disease diagnostics than with therapeutic exploitation due to risk of ubiquitous cellular dysfunction.
