Overview

60S ribosomal protein L34 (RPL34) is a structural component of the large (60S) ribosomal subunit in eukaryotes and is essential for protein synthesis. Encoded by the RPL34 gene, this cytoplasmic protein belongs to the L34E family of ribosomal proteins and contains a zinc finger motif[1][3]. RPL34 has key roles in ribosome assembly and stability and is vital for cell proliferation and viability. Aberrant expression of RPL34 has been observed in multiple human cancers, where it often acts as an oncogene, promoting tumor cell proliferation, migration, and invasion—primarily via engagement of the PI3K/Akt and other growth-related signaling pathways[2][4]. Conversely, in some contexts such as cervical cancer, RPL34 may show tumor suppressor activity by modulating the p53 pathway[4]. Overexpression or mutation can contribute to diseases beyond cancer, such as neurodevelopmental and psychiatric disorders. RPL34 is therefore an emerging target of interest for its diagnostic, prognostic, and therapeutic potential in oncology, but its fundamental cellular function highlights the need for caution in targeting it directly[2][3][4].
