Overview

60S ribosomal protein L36 (RPL36) is a highly conserved structural component of the large (60S) ribosomal subunit, encoded by the human RPL36 gene. It belongs to the L36E family, is located in the cytoplasm, and is essential for ribosome assembly and catalytic protein synthesis. RPL36 also exhibits diverse biological functions beyond structural roles; it promotes cell proliferation and cell cycle progression (notably in glioma), and its alternative protein product (alt-RPL36) modulates cell size and PI3K-AKT-mTOR signaling via interaction with TMEM24. Aberrant expression of RPL36 is observed in hepatocellular carcinoma, where it serves as an independent prognostic marker. Epigenetic mechanisms adjusting RPL36 mRNA stability are involved in cellular responses to toxic insult, such as benzene exposure. Diseases linked to RPL36 include Diamond-Blackfan anemia and pure red-cell aplasia, although direct therapeutic targeting remains unestablished.
