Overview

60S ribosomal protein L9 (RPL9) is a component of the large (60S) subunit of the eukaryotic ribosome and is a member of the L6P family of ribosomal proteins[3][4]. It plays a crucial structural role in the ribosome, participating directly in the process of protein synthesis by catalyzing peptide bond formation and stabilizing the correct folding of rRNA[3][1]. Beyond its canonical ribosomal function, RPL9 has extra-ribosomal activities, including acting as an RNA-binding protein (RBP) that regulates exosomal microRNA (miRNA) cargo and can influence cell proliferation, migration, and invasion in certain cancer contexts, such as hepatocellular carcinoma[2]. Mutations or disruptions in RPL9 are associated with specific diseases, notably Diamond-Blackfan anemia—a congenital erythroid aplasia—and potentially with cancer pathogenesis as an oncogene through its exosomal and miRNA regulatory functions[3][2].
