NIP7 is a nucleolar protein required for accurate and efficient processing of pre-rRNA in eukaryotic cells, primarily functioning in the maturation of the 18S rRNA and assembly of the 40S ribosomal subunit in humans[1][2][3][5][4]. It interacts with other ribosome biogenesis factors such as SBDS and FTSJ3 and is restricted to the nuclear compartment, closely associated with nucleolar pre-ribosomal particles[3][1]. Downregulation of NIP7 affects cell proliferation and ribosome assembly, causing an imbalance in 40S/60S ribosomal subunits and defects in specific pre-rRNA intermediates[1][3]. NIP7 contains a PUA domain responsible for RNA binding, displaying conserved but distinct interactions across archaeal and eukaryotic species[7]. No drugs or clinical biomarkers are associated with NIP7, and it is not a classical therapeutic target[4].
