Overview

8-iso-Prostaglandin F2α (8-iso-PGF2α), also known as 8-isoprostane, is a prostaglandin-like isomer produced in vivo primarily through the non-enzymatic, free radical-catalyzed peroxidation of arachidonic acid [1, 10]. It is widely recognized as the "gold standard" biomarker for assessing oxidative stress and lipid peroxidation due to its chemical stability and presence in various biological fluids like urine and plasma [5, 14]. Beyond its utility as a marker, it is a potent bioactive lipid that exerts significant physiological effects, including intense vasoconstriction and platelet activation [2, 16]. These actions are primarily mediated through its interaction with the Thromboxane A2 (TP) receptor, where it acts as a weak agonist in vascular tissue [3, 7]. Elevated levels of 8-iso-PGF2α are associated with numerous pathological states characterized by oxidative damage, such as cardiovascular disease, hypertension, diabetes, and chronic inflammatory lung diseases [1, 6, 8]. In clinical research, it is used to monitor the efficacy of antioxidant therapies and various medications, such as statins and antihypertensives, which can lower its production by reducing reactive oxygen species [9, 12]. Although not a primary therapeutic target itself, its signaling pathways and production mechanisms are central to the development of treatments for oxidative stress-related disorders [1, 9].
