Overview

Fmoc-Lys-PEG (9-fluorenylmethoxycarbonyl-lysine-polyethylene glycol) and similar nanoparticle core polymers are synthetic materials used as structural scaffolds in drug delivery systems rather than acting as biological targets (Shi et al., 2015, Biomaterials). These amphiphilic molecules consist of a lysine backbone modified with a hydrophobic Fmoc group and a hydrophilic PEG chain, allowing them to self-assemble into nanostructures like micelles or hydrogels (Zheng et al., 2013, Chemical Communications). The primary function of these polymers is to encapsulate therapeutic agents, thereby improving their aqueous solubility and bioavailability. The PEG component provides a "stealth" effect, reducing opsonization and clearance by the mononuclear phagocyte system, which extends the circulation time of the drug (Jokerst et al., 2011, Nanomedicine). In oncology, these nanocarriers exploit the enhanced permeability and retention (EPR) effect to passively accumulate in tumor tissues. While they are not therapeutic targets themselves, they are critical for the formulation of advanced nanomedicines.
