Overview

A disintegrin and metalloproteinase domain-containing protein 10 (ADAM10) is a cell surface transmembrane metalloprotease with a key role in cleaving and releasing the extracellular domains of numerous membrane proteins, including cytokine precursors, cell adhesion molecules, and signaling receptors. It is critical for the proteolytic processing of Notch, amyloid precursor protein (as alpha-secretase), and other substrates important in neural development, immune modulation, and vascular biology. ADAM10 is implicated in multiple diseases, such as cancer, Alzheimer disease, inflammation, and autoimmune disorders, and is under investigation as a therapeutic target, though no drugs specifically targeting ADAM10 are approved for clinical use.
