Overview

A-kinase anchor protein 11 (AKAP11) is a large scaffolding protein belonging to the AKAP family, which functions by binding the regulatory subunits of protein kinase A (PKA) and targeting the holoenzyme to discrete subcellular locations[1][2][9]. AKAP11 is highly expressed in the testis, particularly throughout spermatogenesis and in mature sperm, and it is thought to play roles in cell cycle regulation in both somatic and germ cells as well as in spermatogenesis and sperm function[1][2][6]. AKAP11 interacts with multiple proteins, including GSK3B, protein phosphatase 1 catalytic subunit (PPP1CA), and PKA regulatory subunits, and possibly organizes multi-enzyme signaling complexes[1]. Genetic studies have identified AKAP11 as a risk gene for bipolar disorder[1]. There are currently no approved drugs known to target AKAP11, and it is primarily studied as a scaffolding/signaling regulator rather than a classical drug target[2][9].
