Overview

A-kinase anchor protein 12 (AKAP12), also known as Gravin or SSeCKS, is a large scaffolding protein that plays a critical role in organizing signaling complexes by anchoring protein kinase A (PKA), protein kinase C (PKC), and Src (UniProt: Q02952). It functions primarily as a tumor suppressor, where its loss or epigenetic silencing via promoter hypermethylation is associated with increased cell proliferation, migration, and metastasis in various malignancies, including prostate and breast cancer (PubMed: 15684398, PubMed: 12414643). Beyond its role in oncology, AKAP12 is essential for maintaining the integrity of the blood-brain barrier and regulating vascular dynamics and fibrotic responses (PubMed: 22431538). In the context of drug development, AKAP12 is considered a therapeutic target for reactivation in cancer using epigenetic modifiers like Decitabine, and it is a potential modulator in fibrotic diseases (PubMed: 25623060). Its ability to sequester signaling molecules makes it a critical node in cellular homeostasis and a significant focus for understanding resistance to kinase inhibitors. As a biomarker, the methylation status of the AKAP12 gene is often used to predict disease progression and patient outcomes in clinical research (PubMed: 17322061).
