Overview

A-kinase anchoring protein 10 (AKAP10) is a member of the A-kinase anchoring protein family that binds to the regulatory subunits of protein kinase A (PKA) and positions the holoenzyme to discrete locations within the cell, most notably the mitochondria and plasma membrane. By organizing PKA signaling complexes and anchoring them to specific subcellular compartments, AKAP10 ensures spatial and temporal specificity of cAMP/PKA signal transduction. It interacts with both type I and type II regulatory subunits of PKA and may also facilitate G protein-mediated signaling cascades, acting as a multiprotein complex adaptor. Genetic polymorphisms in AKAP10 have been linked to altered cardiac conduction, arrhythmia risk, and possibly sudden cardiac death, highlighting its critical role in cardiac electrical activity but not as a direct pharmaceutical target at this time.
