A-kinase anchoring protein 3 (AKAP3) is a member of the A-kinase anchoring protein family, serving as a scaffold for protein kinase A (PKA) and other signaling proteins in sperm cells. Expressed specifically in spermatids and mature spermatozoa, AKAP3 is a key component of the sperm tail's fibrous sheath, interacting with other structural proteins such as AKAP4 and partner proteins like CABYR, ROPN1, and SPA17. AKAP3 enables organization of cAMP-dependent PKA signaling complexes that regulate sperm flagellar structure, motility, capacitation, and acrosome reaction. Genetic mutations or disruptions in AKAP3 can lead to male infertility due to compromised sperm motility and improper formation of the fibrous sheath. While AKAP3 is identified as a cancer/testis antigen, it does not currently serve as a direct therapeutic target and has no known interacting drugs[1][2][3][4].
