A-kinase anchoring protein 8 (AKAP8) is a multifunctional nuclear scaffold protein that coordinates several regulatory complexes and processes in the nucleus. It anchors protein kinase A (PKA) and creates spatial microdomains for cAMP-dependent signaling, regulates chromatin architecture by mediating interactions with chromatin-modifying enzymes, and acts directly in the control of gene expression, DNA replication, and mitosis through association with the nuclear matrix and chromatin structures[1][2][3]. As an RNA-binding protein, AKAP8 modulates alternative pre-mRNA splicing events pivotal for maintaining epithelial identity and inhibiting epithelial-mesenchymal transition (EMT), thus suppressing tumor metastasis[2][4]. It also partners with mitotic and chromatin factors such as MCM2 and HDAC3, playing roles in epigenetic regulation and cell cycle progression[2][3]. In cancer, high AKAP8 expression is associated with better prognosis, particularly in breast cancer, where it acts to inhibit EMT and metastasis by regulating alternative splicing of specific transcripts[4]. The function and localization of AKAP8 are modulated by post-translational modifications, including tyrosine phosphorylation by nuclear kinases, leading to changes in chromatin association and gene regulatory activities[1].
