The A20 B-cell lymphoma idiotype refers to the unique antigenic determinant formed by the variable regions of the immunoglobulin heavy and light chains expressed on the surface of A20 cells, a widely used murine model of diffuse large B-cell lymphoma (DLBCL). This idiotype arises from somatic hypermutation (SHM) in IgV genes during B-cell development in germinal centers, producing high-affinity antigen-binding sites.[1] In A20 lymphoma, it functions as part of the B-cell receptor (BCR), enabling chronic antigen-driven signaling that contributes to lymphomagenesis, often alongside NF-κB pathway dysregulation such as A20 (TNFAIP3) inactivation.[1] The idiotype serves as a tumor-specific antigen, potentially targetable for immunotherapy, though no approved drugs are noted; it exemplifies post-germinal center B-cell derivation marked by hypermutated IgV genes.[1][3]
