The A3 adenosine receptor (A3AR), also known as ADORA3, is a G protein-coupled receptor (GPCR) that plays diverse roles in physiological and pathological processes. It is implicated as a therapeutic target for multiple conditions including rheumatoid arthritis, myocardial ischemia, bronchial asthma, and inflammatory diseases. A3AR primarily couples to Gi/Gq proteins, inhibiting adenylyl cyclase activity. Intense activation can induce cell death while low-level activation may be cytoprotective.
Agonists activate A3AR, leading to Gi/Gq protein coupling and inhibition of adenylyl cyclase. Antagonists block A3AR activation.
