AarF domain-containing kinase 1 (ADCK1) is a protein encoded by the ADCK1 gene located on chromosome 14q24.3[1][3][5]. It belongs to the ADCK family, which consists of mitochondrial proteins with putative kinase activity[3][5]. ADCK1 localizes predominantly to the mitochondrial inner membrane and is crucial for regulating mitochondrial dynamics, specifically cristae formation and the balance between mitochondrial fusion and fission[1][3][4][5]. It acts via modulation of mitochondrial structural proteins such as OPA1 and IMMT, with a significant role in ATP production, maintenance of membrane potential, and regulation of reactive oxygen species[2][4][5]. Although ADCK1 is predicted to display serine/threonine kinase activity, most critical functions characterized to date appear to be partially independent of its kinase catalytic activity[4][7]. Pathologically, ADCK1 is upregulated in several cancers, including osteosarcoma and colon cancer, where it promotes tumor cell proliferation and survival via mitochondrial support, and its expression is responsive to mTOR pathway activity[7]. Knockout or silencing of ADCK1 impairs mitochondrial function, leads to increased apoptosis, and can suppress cancer cell growth, making it an emerging therapeutic target in oncology[7]. There are currently no approved drugs that directly target ADCK1, though modulation via mTOR inhibitors has shown utility in preclinical models.
