Overview

AAV receptor (AAVR/KIAA0319L), a cell surface glycoprotein with polycystic kidney disease (PKD) domains, is recognized as the canonical entry receptor for multiple AAV serotypes including AAV8. AAV8 also binds cell surface glycans as attachment factors; in some contexts, the 37/67-kDa laminin receptor (LamR) is strictly necessary for effective AAV8 cellular transduction, in contrast to AAV2, which can use alternative pathways[1][5][6]. The capsid structure of AAV8 determines specificity and efficiency of receptor engagement, shaping tissue tropism and gene delivery outcomes for gene therapies employing AAV8 vectors.
