Abhydrolase domain containing 3 (ABHD3) is an alpha/beta hydrolase fold protein that acts as a lipid-hydrolyzing enzyme (serine hydrolase) with highly selective phospholipase activity for medium-chain and oxidatively truncated phospholipids. It regulates the abundance of specific phospholipids—especially myristoyl (C14)-containing phospholipids—by catalyzing their hydrolysis, thus maintaining cellular lipid homeostasis. ABHD3 is most highly expressed in the brain, small intestine, liver, and kidney, and its regulation is associated with immune response, metabolic balance, and cellular adaptation to stress. Alterations in ABHD3 expression or function have been observed in diseases such as cancer (notably upregulated in chemotherapy response and tumors) and inflammatory conditions like Crohn’s disease. While its full physiological relevance is still under investigation, ABHD3 is recognized as a regulator of phospholipid metabolism, with implications for metabolic, inflammatory, and neoplastic processes.
