Overview

Abhydrolase domain-containing protein 16B (ABHD16B) is an enzyme belonging to the alpha/beta hydrolase domain (ABHD) superfamily. It primarily acts as a lipase with monoacylglycerol lipase and phospholipase activity, capable of hydrolyzing the sn-1 position of glycerophospholipids with high specificity for phosphatidylserine. ABHD16B is involved in regulating cellular lipid homeostasis by affecting genes controlling both neutral lipid degradation (such as triacylglycerol mobilization) and phospholipid synthesis, as supported by experimental overexpression studies. The enzyme modulates lipid droplet dynamics and fatty acid composition, linking it to important physiological processes in lipid metabolism. ABHD16B is expressed in various tissues and across multiple species, where mutations have been correlated with diseases such as Chanarin-Dorfman syndrome, and may affect male fertility by influencing sperm lipid composition. There are currently no known drugs that specifically target ABHD16B, and no information regarding biomarker utility or safety concerns associated with modulation of this target.
