Overview

Abhydrolase domain containing protein 8 (ABHD8) is a member of the alpha/beta hydrolase domain protein family and is predicted to include an alpha/beta hydrolase fold and potential secretory lipase domains[1][5]. The gene is broadly expressed, with notably high levels in testes and brain, but is present in multiple tissues[4][5]. Functional research shows ABHD8 is a scaffold protein that facilitates palmitoylation and subsequent degradation of the NLRP3 protein by promoting chaperone-mediated autophagy, thus negatively regulating inflammasome activation and contributing to immune homeostasis[2]. Genomic studies have implicated ABHD8 in cancer susceptibility, notably breast and ovarian cancer risk, possibly by altering gene expression at the chromosome 19p13.1 locus[1]. There is emerging evidence for a role in mitochondrial regulation, but its direct enzymatic activity, substrate specificity, and complete physiological function remain under investigation[1][4][5]. As a regulatory protein in immune and growth pathways, ABHD8 may represent a candidate therapeutic target for inflammation and certain cancers.
