Overview

Abl interactor 1 (ABI1) is a scaffold and adaptor protein that regulates actin cytoskeletal dynamics and cell signaling. It forms multiprotein complexes, notably with WAVE proteins, Eps8, Nap1, Pir121, and Sos1, facilitating the link between Rac-GTPase activation and Arp2/3-mediated actin nucleation, which is critical for lamellipodia and filopodia formation and cell migration[1][2][3][5][7]. ABI1 is also essential for the assembly of signaling platforms coupling phosphoinositide-3 kinase signaling to Rac activation[1][3]. In neurons, ABI1 is crucial for dendrite and synapse formation, acting as a synapto-nuclear messenger that can influence gene transcription and synaptic plasticity[4]. In smooth muscle and other cell types, ABI1 localizes to the leading edge, promotes F-actin assembly, and interacts with actin regulators such as N-WASP, c-Abl, and profilin-1[7]. Knockdown or disruption of ABI1 impairs cell migration, actin assembly, and normal morphogenesis, with roles implicated in cancer cell invasion, neurodevelopment, and cardiovascular function[3][4][9]. No direct interacting drugs or therapeutic strategies targeting ABI1 are currently described in the literature, and it is not considered a direct therapeutic target, but rather a key modulator in signaling networks and cytoskeletal regulation.
