Acetyl-CoA Carboxylase 2 (ACC2) is a biotin-dependent enzyme localized to the outer mitochondrial membrane. It catalyzes the ATP-dependent carboxylation of acetyl-CoA to malonyl-CoA, a key regulatory step in fatty acid metabolism. ACC2 regulates fatty acid oxidation by producing malonyl-CoA, which inhibits carnitine palmitoyltransferase 1 (CPT1), controlling the entry of long-chain fatty acids into mitochondria for β-oxidation. It is predominantly expressed in heart and skeletal muscle, making it a therapeutic target for obesity, metabolic diseases, and cardiac hypertrophy.
Inhibition of ACC2 to increase fatty acid oxidation by decreasing malonyl-CoA levels, leading to increased CPT1 activity.
