Overview

Acetylcholine biosynthesis refers to the cellular process by which the neurotransmitter acetylcholine (ACh) is produced. The two immediate precursors for acetylcholine synthesis are choline and acetyl coenzyme A (acetyl-CoA). The reaction is catalyzed by choline acetyltransferase (ChAT) in the cytoplasm of nerve terminals. The rate-limiting step is typically the availability of choline. After synthesis, ACh is transported into synaptic vesicles for storage until release upon neuronal stimulation. Once released into the synaptic cleft, ACh acts on its receptors (nicotinic or muscarinic) before being rapidly hydrolyzed by acetylcholinesterase into acetate and choline. The liberated choline is then reabsorbed for reuse in further rounds of synthesis. While not directly targeted, its modulation is crucial for cholinergic neurotransmission, impacting cognitive function and neuromuscular activity.
