Overview

Achaete-scute family bHLH transcription factor 4 (ASCL4) is a member of the basic helix-loop-helix (bHLH) transcription factor family, essential for the determination of cell fate and the development and differentiation of various tissues. ASCL4, also known as HASH4 and BHLHA44, is characterized by relatively restricted gene expression, particularly prominent in fetal skin compared to adult skin, indicating a unique role in skin development. Current knowledge does not support ASCL4 as a direct therapeutic target, and there is no evidence for clinically relevant drug interactions or established biomarker applications. ASCL4 should not be confused with ACSL4, a gene implicated in lipid metabolism, cancer, and ferroptosis, which is sometimes incorrectly abbreviated as "ASCL4" in the literature. The true ASCL4 described here is best classified as a developmental transcription factor with tissue-specific expression and poorly characterized disease relevance in humans[1][4].
