Achaete-scute family bHLH transcription factor 5 (ASCL5) is a member of the achaete-scute complex-like family of basic helix-loop-helix transcription factors. It is a DNA-binding transcription factor predicted to regulate transcription by RNA polymerase II, possibly through sequence-specific binding to E-box motifs in cooperation with other factors such as TCF3/E12. ASCL5 negatively modulates the expression of certain genes, including CDH1/E-cadherin, likely by recruiting the PRC2 repressive chromatin complex to regulatory elements. It plays roles in ameloblast development, tooth germ growth, and enamel formation. While data on ASCL5 remain limited, bioinformatics and gene expression analyses suggest altered expression patterns in cancer: it is upregulated in some lung cancers and downregulated in several brain tumors, implicating a possible role in tumor biology. ASCL5 is part of the broader ASCL transcription factor family, which is critical for development of the nervous system and implicated in progenitor cell biology, injury response, and possibly oncogenesis[1][4][6].
