Overview

Acidic nuclear phosphoprotein 32 family member D (ANP32D) is a protein encoded by the ANP32D gene and is a member of the acidic nuclear phosphoprotein (ANP32) family, which includes proteins that share sequence similarity but have divergent biological roles[1][2][3]. While the related protein PP32 functions as a tumor suppressor, ANP32D is characterized as tumorigenic due to the absence of a 25-amino acid region required for the tumor suppressor activity of PP32[1][2][3]. ANP32D has been implicated in the regulation of cell survival, proliferation, and death, and is also responsive to hypoxic stress, suggesting a role in physiological adaptation to high-altitude environments as well as maladaptive responses such as those seen in chronic mountain sickness[1]. At the molecular level, ANP32D interacts with complexes containing molecular chaperone Hsp90, and its stability may thus be modulated by Hsp90 inhibitors, although no drugs are currently annotated as directly targeting ANP32D[1]. There are no established uses of ANP32D as a therapeutic target, nor are there biomarkers, mechanism-of-action, or safety profiles established for clinical interventions relating to ANP32D at this time[1][2][3].
