Overview

Acrosomal vesicle protein 1 (SP-10; gene symbol ACRV1) is a testis-specific protein that arises during spermatogenesis within the acrosomal vesicle of developing sperm and becomes associated with the acrosomal membranes and matrix of mature sperm[1][5]. Its primary known function is likely related to the binding or penetration of the sperm to the zona pellucida—the glycoprotein layer surrounding the egg—during fertilization, making it integral to male fertility[1]. The ACRV1 gene produces several protein isoforms through alternative splicing and is localized exclusively to sperm, with no evidence of systemic expression[1]. Acrosomal vesicle protein 1 has been studied as a potential immunogen for contraceptive vaccine research, but there is no evidence that it is currently targeted by any approved therapeutic drugs[1][5]. Defects in its expression or function are presumed to contribute to male infertility through impaired acrosome function and fertilization ability[4].
