Overview

Actin-binding LIM protein 1 (ABLIM1) is a multi-domain protein consisting of four LIM domains—protein-protein interaction modules—plus a villin headpiece (VHP) for actin filament binding, and a coiled-coil domain. It mediates the formation and maintenance of cortical actin networks, impacts cell migration, prevents mechanical tension-induced cell blebbing, and acts as a scaffold for signal transduction. As an E3 ubiquitin ligase, it modulates NF-κB signaling by promoting the ubiquitination and degradation of IκBα, thereby activating gene expression programs involved in cancer progression and cell survival. ABLIM1 plays a regulatory role in osteoclastogenesis and bone metabolism via the PI3K/Akt/Rac1 axis. Its expression is tissue-specific, seen notably in retina, brain, muscle, and implicated in several diseases, with contrasting roles in tumorigenesis depending on tumor type.

Mechanism of action

For drugs/inhibitors: targeting its E3 ligase activity could reduce NF-κB activation and downstream oncogenic signals. Targeting its actin-binding function may regulate cell motility and cytoskeletal dynamics.
