Overview

Actin filament-associated protein 1-like 2 (AFAP1L2), commonly known as XB130, is a cytosolic adaptor protein involved in multiple cell signaling pathways, regulating cell proliferation, survival, motility, and gene expression[1][2][3]. It belongs to the adaptor protein family (AFAP family) and functions through protein-protein interactions, containing SH2- and SH3-binding motifs and pleckstrin homology (PH) domains[2][3]. XB130 does not have intrinsic enzymatic activity but interacts with key signaling molecules, such as c-Src and PI3K, influencing PI3K/Akt signaling and downstream survival and cell cycle processes[2][3]. It has prominent roles in cancer biology, with studies identifying its dual nature: promoting tumor progression in some cancer types but correlating with better survival in others[2][3]. XB130 is also implicated in inflammation, airway repair, and broader cytoskeletal organization. Its molecular weight is approximately 130 kDa, and it is highly expressed in thyroid, spleen, and other tissues[2][3]. While it is considered a potential therapeutic and diagnostic target, there are currently no approved drugs known to interact with it directly[1][2][3].
