Overview

Actin-like protein 7A (ACTL7A) is a testis-specific actin-related protein essential for normal sperm head morphology, acrosome formation, and acrosome attachment. It is a structural component of the cytoskeleton and localizes to the acrosome and nucleus of developing male germ cells, playing a critical role in spermatogenesis, fertilization, and early embryonic development. Mutations in ACTL7A are associated with defects in acrosome integrity, impaired fertilization, and male infertility. Beyond its structural roles, ACTL7A and its homolog ACTL7B contribute to the regulation of chromatin structure and gene expression during spermiogenesis, linking nuclear architecture to sperm function and fertility. ACTL7A is not currently recognized as a therapeutic target, and there are no drugs known to modulate its activity.
