Actin maturation protease (ACTMAP, encoded by C19orf54) is an enzyme responsible for removing the N-terminal acetylated methionine from actin after translation, a crucial step for the production of mature actin. Unlike most proteins where initial methionine is cleaved during translation, cytoplasmic actin relies on ACTMAP to excise acetylated methionine post-translationally. Loss or ablation of ACTMAP leads to the accumulation of immature actin across all tissues and prominent defects in skeletal muscle, resulting in shortened actin filaments, reduced muscle function, and classic signs of muscle disease such as centralized nuclei[1][3][5]. There are currently no known drugs or drug mechanisms directly targeting ACTMAP, and there are no established biomarker or safety profiles outside the context of its essential function in actin processing.
