Overview

Actin-related protein 8 (ACTR8, ARP8) is a critical subunit of the evolutionary conserved INO80 chromatin remodeling complex. It contains unique insertions in its actin fold that prevent it from forming filaments, distinguishing it from cytoplasmic actin and ensuring specific recognition of nucleosomes and histone H3–H4 tetramers[1][3][4][5]. As part of the INO80 complex, ACTR8 acts as a nucleosome recognition module, regulating chromatin structure by coupling ATP hydrolysis to nucleosome sliding and repositioning, thereby impacting gene transcription, DNA replication, repair of double-strand breaks, and mitotic chromosome alignment[1][2][3][5]. ACTR8 exhibits low basal ATPase activity and tightly binds ATP, but its structure is locked in a conformation unsuitable for filament formation[3][4]. While ACTR8 is essential for INO80 function and, by extension, maintenance of genome integrity, there is currently no evidence that it serves as a common therapeutic target or biomarker for disease[5].
