Actinidain, also commonly referred to as actinidin, is a cysteine protease enzyme (EC 3.4.22.14) highly abundant in kiwifruit (genus Actinidia), and to a lesser extent present in some other fruits such as pineapple, mango, banana, fig, and papaya. It belongs to the peptidase C1 family (papain-like proteases) and is closely related to enzymes like papain and bromelain, sharing strong similarities in size, structure, and catalytic mechanism. Actinidain is responsible for a substantial portion of the proteolytic activity in kiwifruit, constituting up to 40–50% of the soluble protein content in green (Hayward) kiwifruit at harvest. It exhibits a broad substrate specificity, hydrolyzing a wide range of peptide bonds unattainable by other digestive proteases such as pepsin, and can enhance the digestion of animal and plant proteins in the human gastrointestinal tract. This property underlies its commercial use as a meat tenderizer and as a coagulating agent in dairy product manufacturing. Biochemically, actinidain is active over a wide pH range (pH 3–10; optimum ~pH 5–7), has an optimum temperature around 40°C, and denatures at 60°C—lower than related enzymes like bromelain and papain. The enzyme has been shown to improve the rate and extent of dietary protein breakdown both in vitro and in vivo, suggesting potential as a digestive aid, especially for individuals with reduced digestive efficiency such as the elderly. However, actinidain is also a significant allergen and is the major cause of kiwifruit allergy (allergen listed as Act d 1). Allergic reactions to kiwifruit (mediated by actinidain) can range from mild oral irritation to severe anaphylactic responses. No therapeutic drugs are currently known to specifically target actinidain, and it is not considered a therapeutic target in the sense of a druggable receptor, enzyme, or transporter relevant to disease treatment. Additional notes: - Actinidain is not a human therapeutic target and is not involved in major disease pathways apart from its allergenic potential. - It is often studied as a model enzyme for structure-function analysis among cysteine proteases and as a food ingredient for protein digestion studies. In summary, actinidain is a plant cysteine protease involved in dietary protein digestion and food processing, with significance as a food allergen, but it is not considered a canonical therapeutic drug target.
