Overview

Activating signal cointegrator 1 (TRIP4) is a transcriptional coactivator protein encoded by the TRIP4 gene and functions as a subunit of the tetrameric ASC-1 nuclear complex. TRIP4 interacts with ligand-bound nuclear receptors—including thyroid hormone receptor and retinoid X receptor alpha—to facilitate receptor-mediated transcription in the nucleus. It binds transcriptional coactivators such as EP300, CREBBP, and Nuclear receptor coactivator 1, and also modulates estrogen and androgen receptor activities. TRIP4 participates in the ribosome quality control pathway and is important for neuromuscular and myogenic differentiation. Disease relevance includes its role in spinal muscular atrophy with congenital bone fractures and certain cancers through MAPK, PI3K/AKT, and hTERT signaling activation.
