Activating signal cointegrator 1 complex subunit 2 is a ubiquitin-binding protein encoded by the ASCC2 gene in humans and is a part of the ASCC (Activating Signal Cointegrator 1) complex[2][4][5]. ASCC2 plays a central role in DNA repair, particularly by helping to recruit DNA repair machinery to sites of damage through binding polyubiquitinated proteins bearing Lys-63-linked polyubiquitin chains[2]. It forms part of multi-protein complexes including the ASC-1 and RQT (ribosome quality control trigger) complexes and is involved in resolving stalled ribosomes and in regulating DNA-templated transcription, as well as being implicated in ribosome quality control and protein degradation pathways[1][2]. Somatic mutations in ASCC2 that affect its interaction with ASCC3 have been found in various cancers, highlighting its importance in genome stability and disease[1][3][2]. There are no recorded drug modulators, biomarkers, or notable safety concerns associated with therapeutic targeting of ASCC2 as of current available knowledge.
