Activation of CD4-positive T lymphocytes via antigen presentation by plasmacytoid dendritic cell refers to the process wherein pDCs, a specialized dendritic cell subset, capture and process antigens, then present them on MHC class II molecules to CD4-positive T cells (also known as T helper cells). This interaction, along with co-stimulatory signals (such as CD80/CD86 binding to CD28 on T cells), initiates intracellular signaling cascades leading to T cell activation, proliferation, and differentiation into effector subsets. While conventional dendritic cells are the primary antigen-presenting cells for naive T cell priming, pDCs can also present antigens (although less efficiently) and are best known for secreting high levels of type I interferons, thereby modulating immune responses. This pathway is involved in antiviral defense, autoimmunity, and immune modulation during infection or cancer. Drugs modulating this process typically target individual molecules such as CD4, T cell receptors, co-stimulatory molecules, or cytokines.
