Overview

The activation of CD8-positive cytotoxic T cells (also known as cytotoxic T lymphocytes, or CTLs) is a critical process in the adaptive immune response, enabling the body to target and eliminate cells infected with intracellular pathogens (such as viruses) or transformed by cancer. This activation requires several coordinated signals, including antigen recognition via MHC class I, costimulatory signals (e.g., CD28-CD80/86 interaction), and cytokine support. Once activated, CTLs proliferate, acquire cytolytic capabilities (perforin/granzyme release, Fas/FasL interaction), and secrete cytokines like IFN-gamma. After pathogen clearance, most effector CTLs die, but a subset differentiates into memory populations for long-term immunity. Understanding CTL activation is crucial for vaccine design, immunotherapy, and management of autoimmune and transplant-related disorders.
