Overview

Activator protein 1 (AP-1) is a transcription factor complex that regulates gene expression in response to a wide range of stimuli. It is a dimeric complex formed by members of the Jun, Fos, ATF, Maf, and JDP protein families. AP-1 plays critical roles in cellular processes such as differentiation, proliferation, apoptosis, and oncogenic transformation. Activated AP-1 binds to specific DNA sequences (TREs and CREs) in promoter regions of target genes. Dysregulation of AP-1 has been implicated in cancer development/progression due to its role in cell cycle control and apoptosis resistance.

Mechanism of action

Modulation of AP-1 activity often involves targeting upstream kinases in the MAPK and PI3K/Akt pathways, or direct inhibition of AP-1 dimerization or DNA binding.
