Activator protein 1 (AP-1) is a dimeric transcription factor complex composed mainly of proteins from the Fos, Jun, ATF, and JDP families. It regulates gene expression in response to various stimuli, including cytokines, growth factors, stress, and pathogens. AP-1 controls crucial cellular processes such as proliferation, differentiation, apoptosis, and senescence. Dysregulation of AP-1 activity is implicated in cancer, inflammation, and other diseases, making it an attractive but challenging therapeutic target. Several small-molecule and natural compound inhibitors of AP-1 have been identified, but clinical efficacy remains unproven due to the complexity and ubiquity of AP-1 signaling[1][2][4][5][6].
