Overview

The Activator protein 1 (AP-1) transcription factor complex is a dimeric protein assembly, most commonly formed by the heterodimerization of c-Fos and c-Jun subunits. It belongs to the basic leucine zipper (bZIP) family and binds to the TPA-responsive element (TRE) in the promoter regions of target genes. AP-1 acts as a central integrator of various signaling pathways, including the MAPK/ERK and JNK pathways, regulating critical cellular processes such as proliferation, differentiation, and apoptosis (UniProt P01100; PubMed: 11907574). In pathological states, particularly cancer and chronic inflammation, AP-1 is frequently overexpressed or hyperactivated, driving the transcription of genes involved in tumor invasion, metastasis, and the production of pro-inflammatory cytokines (PubMed: 15928673). Therapeutic targeting of the c-Fos/AP-1 complex has focused on small molecules like T-5224, which selectively inhibit its DNA-binding activity to treat conditions like rheumatoid arthritis and various malignancies (PubMed: 18414293). Despite its potential, the broad biological influence of AP-1 poses challenges for achieving therapeutic selectivity without significant side effects.
