Activins are dimeric proteins belonging to the TGF-β superfamily, crucial for regulating various processes including embryonic development, cell proliferation and differentiation, immune responses, and reproductive system function. They signal through type II and type I serine/threonine kinase receptors, activating SMAD transcription factors. Activin activity is tightly controlled by inhibitors like follistatin.
Activin binds to type II receptors (ActRIIA/B), which then recruit and phosphorylate type I receptors (ALK4/ALK7). Activated type I receptors phosphorylate SMAD2/SMAD3 proteins, leading to gene transcription.
