Activin receptor type IIB (ActRIIB), encoded by the ACVR2B gene, is a transmembrane serine/threonine kinase receptor that belongs to the transforming growth factor-beta (TGF-β) superfamily of receptors. It plays a central role in mediating signals from ligands such as activins and myostatin, which are involved in regulating cell growth, differentiation, and various physiological processes. Blocking ActRIIB or its interaction with ligands like myostatin has been explored as a therapeutic strategy for conditions involving muscle wasting.
Ligand binding induces formation of a heteromeric complex with type I receptors, leading to phosphorylation of SMAD proteins and regulation of gene transcription. Blocking ActRIIB or its interaction with ligands like myostatin inhibits signaling.
