Acyl-CoA Cholesterol Acyltransferase 1 (ACAT1) is an enzyme that plays a crucial role in lipid metabolism. It catalyzes the formation of acetoacetyl-CoA from two molecules of acetyl-CoA, a key step in ketogenesis and ketolysis. ACAT1 also participates in isoleucine degradation and the esterification of cholesterol with long-chain fatty acyl CoAs to form cholesteryl esters. It is implicated in beta-ketothiolase deficiency and is a potential therapeutic target in cancer due to its role in glycolysis and tumor growth.
