Overview

ACSL5 is an isozyme of the long-chain fatty-acid-coenzyme A ligase family. It catalyzes the conversion of free long-chain fatty acids (C16–C20) into their active form, fatty acyl-CoA esters. This activation is essential for both lipid biosynthesis and degradation via β-oxidation. The enzyme plays a pleiotropic role in lipid metabolism depending on substrate preferences, subcellular localization, and tissue specificity.
