Overview

Acyl-CoA thioesterase 7 (ACOT7) is a lipid-metabolizing enzyme predominantly expressed in the brain and some immune and cancer cells. It catalyzes the hydrolysis of long-chain acyl-CoA esters to free fatty acids and coenzyme A, regulating cellular lipid and energy metabolism. ACOT7 protects neurons from lipotoxicity, is critical for myelin development and maintenance, and has a strong role in eicosanoid (prostaglandin) metabolism. Overexpression is associated with poor prognosis in various cancers and correlates with cell proliferation and immune microenvironment modulation. In Alzheimer’s disease, elevated serum ACOT7 correlates with disease severity and is a promising noninvasive biomarker, potentially influencing amyloid precursor protein metabolism and amyloidogenic processing. Its expression patterns and regulatory impact on lipid intermediates make it a potential therapeutic target in neurodegeneration, cancer, and inflammation, though drug development may be challenged by necessary physiological functions in brain and metabolic tissues.
