Overview

ADAM metallopeptidase domain 29 (ADAM29) is a membrane-anchored protein and a member of the ADAM (A Disintegrin And Metalloproteinase) family, structurally related to snake venom disintegrins. While all ADAMs contain a metalloproteinase domain, ADAM29 appears to be catalytically inactive (i.e., it is a non-catalytic metalloprotease-like protein) due to mutations in the active site, suggesting its biological role is primarily via protein-protein interactions rather than enzymatic proteolysis. It is highly expressed in the testis and may play a role in spermatogenesis and fertilization by facilitating cell-cell or cell-matrix interactions required in these processes. Dysregulation of ADAM29 has been linked to diseases such as melanoma, but, unlike better studied ADAM family members, there are currently no drugs or specific mechanisms of action reported for it, nor is it established as a clinical biomarker or safety concern.
