Overview

ADAM metallopeptidase domain 3A (ADAM3A) belongs to the ADAM (a disintegrin and metalloproteinase) family, which typically have roles in cell adhesion, migration, proteolysis, and signaling[1][3]. However, ADAM3A is annotated as a pseudogene in humans and does not produce a functional protein[6]. Some other ADAM family members are important proteases involved in ectodomain shedding, reproductive biology (notably in rodents), and disease; however, these functions are not applicable to ADAM3A in humans. Reports regarding ADAM3A typically refer to data from non-human species (e.g., rodents or porcine studies) or are confounded by naming conventions; for example, in rats and pigs, ADAM3a may code for functional proteins involved in sperm-egg interactions[4][5], but this does not apply to humans. In short, ADAM3A is a pseudogene in humans, is not a therapeutic target, and does not have biological or clinical relevance[6]. Any disease associations, drug interactions, or molecular functions attributed to ADAM3A are misplaced, and care should be taken to avoid conflating this "target" with other functional ADAM family members.
