Overview

ADAMTS-like protein 5 (ADAMTSL5) is a secreted, N-glycosylated glycoprotein of the ADAMTS-like family found in the extracellular matrix and on cell surfaces[1][2][3]. It is characterized by a unique modular organization including an N-terminal thrombospondin type-1 repeat, a cysteine-rich and spacer module, and a C-terminal netrin-like (NTR) module. ADAMTSL5 binds to both fibrillin-1 and fibrillin-2, components of microfibrils in the extracellular matrix, implicating it in the regulation of microfibril function and ECM architecture[1][2][3]. It is broadly expressed in musculoskeletal and epithelial tissues. In the immune context, ADAMTSL5 is a melanocytic autoantigen presented by HLA-C*06:02, activating autoreactive T cells and contributing to psoriatic inflammation; its autoantibodies correlate with disease severity in psoriasis and psoriatic arthritis, highlighting a role as a potential biomarker[2]. No known drugs selectively target ADAMTSL5 and it is not currently considered a therapeutic target[3].
