Overview

ADAMTS9 antisense RNA 2 (ADAMTS9-AS2) is a long non-coding RNA located on chromosome 3q26.2, transcribed antisense to the ADAMTS9 gene[2]. As a long non-coding RNA, ADAMTS9-AS2 does not encode a protein but regulates gene expression, notably by acting as a competing endogenous RNA (ceRNA) and sponging specific microRNAs such as miR-942-5p and miR-153-5p[1]. It is involved in cartilage regeneration by promoting chondrogenic differentiation of human mesenchymal stem cells (hMSCs) via upregulation of key chondrogenic genes and the transcription factor Scrg1[1]. ADAMTS9-AS2 can also modulate the expression of its sense gene, ADAMTS9, thereby influencing tissue homeostasis and extracellular matrix remodeling[2][4]. Dysregulation or altered expression of ADAMTS9-AS2 has been implicated in several diseases, notably various cancers, rheumatoid arthritis, and osteoarthritis[2][3][4]. In cancer, ADAMTS9-AS2 may suppress tumorigenesis and metastasis through inhibition of signaling pathways such as PI3K/AKT/mTOR and by modulating target genes involved in cell cycle regulation and apoptosis[3][4]. Its lower or higher expression in pathological contexts underscores its potential utility as a biomarker, but there are currently no approved drugs that directly target or utilize ADAMTS9-AS2[3][4].
