Overview

ADAMTSL4 antisense RNA 2 (ADAMTSL4-AS2) is a long non-coding RNA (lncRNA) gene located on chromosome 1, transcribed antisense to the protein-coding gene ADAMTSL4. It belongs to the class of antisense lncRNAs, which are known to regulate gene expression of their sense counterparts. Experimental evidence suggests ADAMTSL4-AS2 may influence ADAMTSL4 expression via interactions with the enhancer and promoter regions of the sense gene, positively regulating its transcription[2]. ADAMTSL4-AS2 has been implicated as a gene associated with ocular phenotypes such as ectopia lentis and craniosynostosis with ectopia lentis, likely through its regulatory role on ADAMTSL4, a gene critical for extracellular matrix structure in the eye[6]. However, there is no direct evidence it is a therapeutic target, nor are there known small-molecule drugs or biologicals in clinical use or investigation that target this RNA[1][3][6]. This target is not a receptor, enzyme, or protein but is an lncRNA gene, so it should not be considered a classical therapeutic target. ADAMTSL4-AS2 should be distinguished from its sense gene, ADAMTSL4, which is a protein-coding gene and causally implicated in ocular diseases.
